Isotype controls are useful in discriminating signal from noise in flow cytometry experiments. Isotype control antibodies are not specific for any antigens on the target cell and represent the background levels of non-specific fluorescence associated with cells after they have been stained with fluorochrome conjugated antibodies.
A proper isotype control will meet the following three criteria:
- It will be of the same host species as the primary antibody for which it is a control.
- It will be conjugated to the same fluorochrome as the primary antibody for which it is a control.
- It will be used at the same concentration as the primary antibody for which it is a control.
